Cutinase can be evaluated for hydrolysis of cutin and selected ester-containing materials. A useful materials trial distinguishes a surface modification from substantial polymer breakdown and does not assume every polyester is equally susceptible.

Describe the material as well as the enzyme

Record polymer identity, film thickness, crystallinity where available, surface area and prior treatment. Cutin is the defining natural substrate, but performance on a commercial plastic depends on its structure and accessibility. A successful result with an easily hydrolysed ester assay substrate does not establish useful degradation of a PET article or another resistant polymer.

Choose the claim the experiment can support

Contact-angle change can indicate altered surface wettability, but it does not quantify bulk depolymerisation. For breakdown, measure released products, molecular changes and appropriately washed, dried mass. Include a no-enzyme control exposed to the same temperature, pH and agitation. Additives or loosely bound material may leach without polymer-chain conversion.

Measure activity and material outcomes separately

Test a suitable reference substrate to check enzyme survival during the chosen hold. Use the supplied grade’s actual stability window; high-temperature polymer research may use engineered enzymes that are not equivalent to the commercial preparation. Analyse the surface and liquid products together, then assess whether the modified material retains the mechanical or functional properties required downstream.

Troubleshooting

  • Mass changes in both control and treatment: assess leaching and washing losses.
  • Reference activity persists but polymer is unchanged: investigate polymer accessibility and specificity.

Common questions

Does a lower contact angle prove plastic recycling?

No. Surface wetting and bulk conversion are different endpoints.

Will a paper about an engineered cutinase predict this grade?

Not automatically. Compare the enzyme identity and process conditions.
