Investigate activity loss by comparing a retained reference with the suspect sample using the same assay. Storage temperature, dilution, handling and formulation exposure can all matter, but an apparent loss can also be caused by an inconsistent test.

Start with an interpretable comparison

Use the same substrate batch, assay method, pH, temperature and sampling interval. Include blanks and confirm the assay remains within its useful measurement range. An old result from a different method is a weak baseline for deciding that a stored preparation has deteriorated.

Reconstruct the handling history

Record receipt date, lot, opening date, storage conditions, transport excursions and any freezing or heating. Follow the product-specific instructions. There is no single storage rule appropriate to every liquid, powder, immobilised or formulated enzyme preparation.

Treat a dilution as a new formulation

Dilution changes the concentration of stabilising ingredients as well as the enzyme. The unopened concentrate shelf-life statement does not establish the stability of a diluted working solution. Prepare and validate working solutions for the intended holding period and prevent contamination during handling.

Separate storage loss from process inhibition

If the preparation is active in its reference assay but weak in the application, examine the process matrix and conditions. If a stored detergent loses cleaning performance, test the complete formulation as well as retained enzyme activity. Loss of performance cannot always be attributed to the enzyme alone.

Should every enzyme be refrigerated?

Follow its specific storage instructions; a universal rule may be inappropriate for the formulation.

Does freezing preserve every enzyme?

No. Assess freezing and thawing only where supported for the particular preparation and intended use.

  • The reference and suspect samples both test weak: investigate the assay before concluding storage loss.
  • Concentrate is stable but its dilution declines: assess diluent, cleanliness and the working-solution hold.
