96-Well β-Glucosidase Activity Assay Using pNPG

Why is this done?

β-Glucosidase releases glucose from short cellulose-derived sugars; a pNPG assay gives a fast, sensitive proxy for comparing preparations. Activity on pNPG does not automatically predict activity on cellobiose or biomass.

How does it work?

Let the enzyme release yellow p-nitrophenol from pNPG, stop the reaction at a fixed time, and compare A₄₀₅ with standards treated the same way. Use blanks and multiple dilutions to identify the linear region.

Materials and equipment

  • pNPG substrate, pNP reference standard, assay buffer suited to the grade, and 1 M sodium carbonate stop solution.
  • 96-well reaction plate suitable for the incubation temperature, clear readout plate and reader at 405 nm.
  • A dilution series of enzyme and matched buffer, substrate and sample blanks.

Procedure

  1. Prepare a pNP standard series in the same final stopped-reaction matrix. For an illustrative 100 µL reaction, combine 50 µL assay buffer, 25 µL pNPG stock and 25 µL diluted enzyme; choose substrate stock so the final concentration is 1–5 mM. Run at least two enzyme dilutions in duplicate or triplicate.
  2. Pre-equilibrate buffer and substrate to the chosen temperature. Start timed columns by adding enzyme, mix consistently, and incubate for a selected short linear interval such as 10 minutes at 37°C. Check linearity at 5, 10 and 15 minutes on the first run.
  3. Stop each well in the same order with a fixed volume of 1 M sodium carbonate, such as 100 µL for a 100 µL reaction. Treat pNP standards and blanks identically. Transfer to a clear plate if the incubation plate is not optically suitable, then read A405 promptly.
  4. Use the pNP calibration in nmol per well to obtain corrected product formed. Select wells within both calibration range and time/enzyme linear range.

Controls and blanks

Include enzyme plus no substrate, substrate plus no enzyme, and a heat-inactivated or buffer control. Coloured crude samples require sample blanks; some formulations can contribute pNP-like absorbance.

Calculations and reporting

Activity in U/mL of original enzyme = pNP formed (nmol) × original dilution factor / [reaction time (min) × enzyme volume (µL)]. This equality follows because 1 U = 1 µmol/min and 1 mL = 1000 µL. For 50 nmol in 10 min from 25 µL of a 1:100 dilution, activity is 20 U/mL. State the pH, temperature and substrate concentration.

Quality checks and troubleshooting

If A405 is nonlinear with time or dilution, shorten the incubation or dilute the enzyme. pNP absorbance depends strongly on pH; standards must see the same carbonate and final volume. Compare substrate-specific pNPG units only with assays under matched conditions.

Method scope

This is a research and process-development protocol. Verify the selected enzyme grade, reagent supplier instructions, equipment capacity and local laboratory SOP before use. Report the exact assay definition and conditions; results obtained with different substrates or methods are not automatically interchangeable.

Illustrative pNP calibration dataset

Illustrative, simulated values for graphing and calculation practice. These are not measured STL product data or acceptance limits.

x-axis: final p-nitrophenol concentration in stopped well (µM); y-axis: blank-corrected A₄₀₅ (dimensionless absorbance).

Data pairs (x → y): 0→0.000; 20→0.120; 40→0.240; 60→0.360; 80→0.480; 100→0.600.

Simulated y = 0.0060x, with x in µM. An assay at A₄₀₅ = 0.300 gives 50 µM in a 200 µL stopped well, or 10 nmol pNP. Report U/mL using incubation time and volume of enzyme added, then multiply by sample dilution; pNP standards need the same pH and final volume.
