Acid protease
Select an acid-active protease for low-pH protein processing, fermentation or a defined research digestion.
01 / Understand the chemistry
How does acid protease work?
Acid protease is an application category. The representative example is aspergillopepsin I, an aspartic endopeptidase. Catalytic mechanism, substrate preference and stability must be established for the actual preparation.

03 / Methods & measurement
How to measure acid protease activity
Use a protein substrate validated at the assay pH, with blanks for pH-dependent precipitation and absorbance. Compare hydrolysates using an appropriate peptide or amino-group method.
Make a better selection
What matters when choosing a grade?
Acidification alone can change protein solubility. Apparent clarification or precipitation is not proof that peptide-bond hydrolysis occurred.
Confirm source, grade, activity method, formulation, storage and compatibility. Use stability data for the selected preparation; an optimum measured for one protein does not define every product’s operating window.
From screening to production
Plan a useful acid protease trial
Set a matched control
Use your actual material. Keep pH, temperature, residence time and mixing identical in the untreated control, so physical processing effects are separated from enzyme action.
Screen dose and time
Compare a dose ladder and take a time course. State activity units and their assay definition, and express dose against a clear feedstock or process basis.
Validate the endpoint
Measure solubility, hydrolysis, bitterness and yield. Assess retained activity, downstream quality and cost per useful outcome before increasing scale.

Acid protease: common selection questions
What does acid protease produce?
Acid protease is an application category. The representative example is aspergillopepsin I, an aspartic endopeptidase. Catalytic mechanism, substrate preference and stability must be established for the actual preparation.
How do I test acid protease for my process?
Use a protein substrate validated at the assay pH, with blanks for pH-dependent precipitation and absorbance. Compare hydrolysates using an appropriate peptide or amino-group method.
What should I check before buying acid protease?
Acidification alone can change protein solubility. Apparent clarification or precipitation is not proof that peptide-bond hydrolysis occurred. Share your feedstock, operating conditions, target outcome and required documentation when requesting a grade recommendation.
