Alkaline protease
Develop protein-stain removal or controlled protein hydrolysis using a protease compatible with the actual alkaline formulation.
01 / Understand the chemistry
How does alkaline protease work?
Alkaline protease is a process description, not a single EC class. This page uses subtilisin, a serine endopeptidase, as its representative example. Different preparations have different cleavage preferences, stability and formulation tolerances.
Expose the protein
Hydration and substrate unfolding can make peptide bonds accessible.

How to measure alkaline protease activity
Casein, azocasein or a defined peptide substrate can support activity testing. State the substrate and unit definition; units from different protease methods are not automatically comparable.
Make a better selection
What matters when choosing a grade?
Proteases can digest other enzymes in a blend. Compatibility and retained activity during storage must be tested in the complete formulation.
Confirm source, grade, activity method, formulation, storage and compatibility. Use stability data for the selected preparation; an optimum measured for one protein does not define every product’s operating window.
From screening to production
Plan a useful alkaline protease trial
Set a matched control
Use your actual material. Keep pH, temperature, residence time and mixing identical in the untreated control, so physical processing effects are separated from enzyme action.
Screen dose and time
Compare a dose ladder and take a time course. State activity units and their assay definition, and express dose against a clear feedstock or process basis.
Validate the endpoint
Measure stain removal and fabric compatibility in the finished wash system. Assess retained activity, downstream quality and cost per useful outcome before increasing scale.

How do I test alkaline protease for my process?
