Lipase
Select lipase for fat hydrolysis, oil modification or a defined ester synthesis. Water activity and the reaction medium strongly affect the outcome.
01 / Understand the chemistry
How does lipase work?
Triacylglycerol lipases hydrolyse ester bonds in glycerides. In suitable low-water systems, selected lipases can also catalyse esterification or transesterification. The CALB structural example illustrates one protein, not every industrial lipase.

How to measure lipase activity
A p-nitrophenyl ester assay is a convenient screen, but performance can differ on real oils. Use titration, chromatography or a substrate-specific method to confirm the target reaction.
Make a better selection
What matters when choosing a grade?
Water is a reactant in hydrolysis but a key process variable in synthesis. An aqueous assay ranking may not predict conversion in a low-water oil system.
Confirm source, grade, activity method, formulation, storage and compatibility. Use stability data for the selected preparation; an optimum measured for one protein does not define every product’s operating window.
From screening to production
Plan a useful lipase trial
Set a matched control
Use your actual material. Keep pH, temperature, residence time and mixing identical in the untreated control, so physical processing effects are separated from enzyme action.
Screen dose and time
Compare a dose ladder and take a time course. State activity units and their assay definition, and express dose against a clear feedstock or process basis.
Validate the endpoint
Measure FAME by a validated chromatographic method and assess product quality. Assess retained activity, downstream quality and cost per useful outcome before increasing scale.

Lipase: common selection questions
What does lipase produce?
Triacylglycerol lipases hydrolyse ester bonds in glycerides. In suitable low-water systems, selected lipases can also catalyse esterification or transesterification. The CALB structural example illustrates one protein, not every industrial lipase.
How do I test lipase for my process?
A p-nitrophenyl ester assay is a convenient screen, but performance can differ on real oils. Use titration, chromatography or a substrate-specific method to confirm the target reaction.
What should I check before buying lipase?
Water is a reactant in hydrolysis but a key process variable in synthesis. An aqueous assay ranking may not predict conversion in a low-water oil system. Share your feedstock, operating conditions, target outcome and required documentation when requesting a grade recommendation.
