Pectinase

Release value from pectin-rich materials.

Choose a pectinase system for juice processing, plant extraction or pectin-rich waste conversion. The name describes a group of activities, not one universal enzyme.

SUBSTRATEPectin / polygalacturonate
REACTIONα-1,4 galacturonosidic bonds
PRINCIPAL OUTPUTShorter galacturonan chains
EC EXAMPLE3.2.1.15

01 / Understand the chemistry

How does pectinase work?

The structural and EC example here is endo-polygalacturonase, which hydrolyses the galacturonan backbone. Pectin lyases, pectate lyases and pectin methylesterases perform different reactions and can be relevant to the same feedstock.

Identify the pectin

Degree of methylesterification and plant origin affect the appropriate enzyme system.

MEASURE SUCCESS

Measure soluble carbon, galacturonic acid and downstream performance.

PROCESS DECISION

Balance pectin chemistry with the other feedstock polymers.

03 / Methods & measurement

How to measure pectinase activity

Use a defined polygalacturonic-acid assay for polygalacturonase, and measure the process endpoint separately. Lyase assays and methylesterase assays do not measure the same reaction.

Make a better selection

What matters when choosing a grade?

A pectin methylesterase step can release methanol from methylesterified pectin. Enzyme composition matters when selecting a preparation for food and beverage processing.

Confirm source, grade, activity method, formulation, storage and compatibility. Use stability data for the selected preparation; an optimum measured for one protein does not define every product’s operating window.

From screening to production

Plan a useful pectinase trial

Set a matched control

Use your actual material. Keep pH, temperature, residence time and mixing identical in the untreated control, so physical processing effects are separated from enzyme action.

Screen dose and time

Compare a dose ladder and take a time course. State activity units and their assay definition, and express dose against a clear feedstock or process basis.

Validate the endpoint

Measure turbidity, juice yield, viscosity and filterability. Assess retained activity, downstream quality and cost per useful outcome before increasing scale.

Pectinase: common selection questions

What does pectinase produce?

The structural and EC example here is endo-polygalacturonase, which hydrolyses the galacturonan backbone. Pectin lyases, pectate lyases and pectin methylesterases perform different reactions and can be relevant to the same feedstock.

How do I test pectinase for my process?

Use a defined polygalacturonic-acid assay for polygalacturonase, and measure the process endpoint separately. Lyase assays and methylesterase assays do not measure the same reaction.

What should I check before buying pectinase?

A pectin methylesterase step can release methanol from methylesterified pectin. Enzyme composition matters when selecting a preparation for food and beverage processing. Share your feedstock, operating conditions, target outcome and required documentation when requesting a grade recommendation.
